Honestly, I am not the right person to answer that, cuz I don't really use anti-spyware or anti-virus.
I just use it with care. But I know that AVG is a pretty popular software for anti-spyware.
I used a program called hijackthis, it is not an anti-spyware program, but a lot of people like to use it to troubleshoot potential spyware infected PC's. It lists all the programs that your PC launches at startup, all the browser helper objects (BHO), and some other things that will give you a hint on what has been running on your computer (some of them are good, but some of them might be bad). And then if you see something unusual, you can do some research on the web about that, and decide whether you want to clean it up (manually). For example, if you see that it runs a file called xyz.exe on startup, and you don't recall installing that program, you check on the web to learn about that file, and if you found that xyz.exe is an known spyware process or some security thread, then you can clean it so it will not launch it, and delete the file manually. WARNING: if you don't know what you are doing, you can mess up your computer.
